Product for the preparation of cyto-histological samples for optical microscopy. Method for the demonstration of spirochetes.
The argyrophilic reaction is based on the capacity of bacteria cell wall to reduce the silver on its metallic state binding them in a visible form.
METHOD:
1) Bring section to distilled water.
2) Preparation of ā€˜impregnating-solutionā€™: place the slides container in the polystyrene support. Introduce in the container 13 ml of distilled water; add 4,5 ml of reagent A and 20 drops of reagent B. Shake briefly with a glass stick previously washed in distilled water.
3) Introduce the slide in the solution and incubate for 90 minutes at 60-70Ā°C.
4) Take out the holder from oven and let it cool for 5 minutes.
5) During the impregnating reaction prepare the ā€˜developing solutionā€™. VERY IMPORTANT: it is advisable to prepare the solution during the last 12 minutes of incubation started at point 3). Introduce 6 ml of reagent C in in the second slide holder. Preheat in the oven at 50Ā°C for 10 minutes the reagent C and a bottle D.
6) Add the reagent D at reagent C in the second slide holder. Shake briefly with a glass stick previously washed in distilled water, then introduce 5 ml of reagent E and shake again.
7) Introduce the slide in the ā€˜developing solutionā€™ and put in the oven at 50Ā°C for 5-10 minutes.
8) Wash in running tap warm water for 2 minutes.
9) Dehydrate through ascending alcohols: xylene and mount.
